Cloning and Sequencing of a Putative Protease Gene from Burkholderia Pseudomallei
註釋Burkholderia pseudomallei is the causative agent of melioidosis which is endemic in South East Asia and northern Australia. This Gram-negative bacillus has been known to produce a variety of extracellular proteins believed to play a role in its pathogenicity. Amongst these virulence factors is the extracellular protease. The protease was reported to be a metalloprotease with a molecular weight of 36 kDa. This protease was reported to attack protein of the immune system and is also capable of causing dermonecrosis and corneal damage. A library of putative B. pseudomallei protease gene was constructed in Escherichia coli JM109 using pQE-40 and pLITMUS-28 expression vectors.
