Polyadenylation of Messenger RNA and Termination of Transcription in Polyomavirus [microform]
註釋Circumstantial evidence suggested that polyadenylation and termination of transcription may be linked. To test this hypothesis, we constructed viable insertion mutants of polyomavirus that contain a second functional polyadenylation signal. We showed that the polyadenylation of L-strand RNAs requires sequence elements located between 12 and 87 nucleotides downstream of AAUAAA. Polyadenylation ofE-strand RNAs requires no more than 19 nucleotides upstream and 44 nucleotides downstream of AAUAAA. An insertion mutant (ins 4) that contained a second functional L-strand signal produced L-strand RNAs with 3' ends at both sites. However, the efficiency of polyadenylation and that of transcription termination remained as low as that in wild type-infected cells. [...].
