Induction of Osteogenic Differentiation in Human Mesenchymal Stem Cells by Crosstalk with Osteoblasts
註釋Zusammenfassung: Natural bone healing following fractures is initiated by osteoblasts (OB), mesenchymal stromal cells (MSC), and chondrocytes, a cell combination with possible potential in tissue engineering techniques for bony defects. The aim of the study was to investigate crosstalk among OB, MSC and chondrocytes, in order to determine optimal cell culture conditions for osteogenic differentiation. Human cell interactions were investigated in an in vitro trans-well co-culture study over a time period of 28 days by measurement of the OD (optical density) at 450nm. According to the stages of osteogenesis proliferation, specific matrix production and calcification were assessed. Proliferation was measured by the Cell Titer Blue assay. Specific matrix production was determined by quantitative PCR. Calcification was assessed through gene expression of ALP and Alizarin Red staining. Chodrogenesis was quantified by the DMMB assay. We found that OB can induce MSC to differentiate along the osteogenic pathway by reducing proliferation, increasing specific matrix production and increasing calcification.
