By a shotgun cloning approach, a putative suppressor tRNA capable of altering a proline auxotroph of Escherichia coli to proline prototrophy was choled from the genomic DNA of Thermus ruber, a moderately thermophilic bacterium. By the same approach, hybrid kanamycin resistance plasmids harbouring a thermostable kanamycin adenyltransferase gene from pKT1 plasmid and the replication origin of Thermus aquaticus YT-1 or Thermus thermophilis HB8 were constructed in T. thermophilus HB27.
